— MoxiFlow(圖1)。

對于它外觀的小巧此處不再累述(大小規(guī)格僅為26×15×15cm),圖片比文字更有說服力(圖2);儀器內(nèi)置4500mAh鋰電池,便于在實驗室內(nèi)的移動和隨意放置;內(nèi)置軟件程序,480X320pix彩色顯示器可清晰呈現(xiàn)數(shù)據(jù)結(jié)果,讓您一目了然。至于檢測方式,它采用了兩種檢測的**融合—庫爾特電阻原理與熒光檢測原理:

一、 庫爾特原理(用于檢測細胞大小及數(shù)量)
當(dāng)懸浮于液體中的細胞通過小孔管時,會取代相同體積的液體,在恒定電流設(shè)計的電路中導(dǎo)致小孔管內(nèi)外的電阻發(fā)生瞬時變化,產(chǎn)生電位脈沖,而這種脈沖信號的大小和次數(shù)與細胞的大小與數(shù)目成正比,因此可用于細胞計數(shù)及大小的檢測。這正是談到流式細胞儀的歷史而不可避開的庫爾特原理,庫爾特電阻檢測法屬于對細胞個體的測量和三維的測量,不但能準(zhǔn)確測量細胞的大小,更能對細胞的**數(shù)量及濃度進行測量。利用庫爾特原理檢測細胞大小更近真實,而不像激光衍射散射原理會受到染料顏色和濃度的影響。流式細胞儀(FCM,Flow
cytometry),從字面上的cytometry
(細胞計數(shù)) 也看得出它與細胞計數(shù)有著一定的淵源,Wallace
Coulter于1947年發(fā)明了庫爾特原理,用于血細胞分析,隨后1953年Coulter公司便推出了世界上**臺流式細胞分析儀。而以庫爾特原理為主導(dǎo)技術(shù)開發(fā)細胞計數(shù)儀的ORFLO公司在此推出這樣一臺眾望所歸的微型流式細胞儀也便是名正言順了,并利用其**的薄膜傳感器技術(shù)對細胞進行數(shù)量、直徑等檢測,獲取更加精準(zhǔn)可靠的數(shù)據(jù)。
二、 熒光檢測(用于檢測熒光信號)
MoxiFlow配置了一根532nm固態(tài)激光器和一個PMT,檢測光譜區(qū)域為590/40nm
( 可檢測染料:R-PE,
PI, Nile Red,
Ethidium Homodimer I & III, Sytox Orange等 )。對細胞大小檢測的同時以傳統(tǒng)流式的方法利用激光激發(fā)標(biāo)記在細胞上的特異性熒光染料,并通過PMT進行檢測,獲取熒光信號。
MoxiFlow 即利用了以上兩種檢測方式相結(jié)合的方式,在細胞流經(jīng)小孔管時,同時對細胞的大小、數(shù)目及熒光信號進行**的檢測(如圖3)。
化 繁 為 簡
一、 簡易化操作
微流體系膜片內(nèi)設(shè)細胞過濾功能,所以對于準(zhǔn)備上機的細胞懸液無需進行濾網(wǎng)的過濾,直接即可加入膜片孔內(nèi)準(zhǔn)備實驗(如圖4),此外膜片的窗口處為非透明材質(zhì)做成,形成小的避光暗室,避免了熒光檢測過程中熒光猝滅現(xiàn)象;僅需兩步即可完成所有操作步驟。一,將細胞微流膜片插入檢測口;二,將50μL樣品加入檢測孔內(nèi),點擊Play即可開始檢測 (如圖5~6);僅10秒鐘即可顯示出實驗結(jié)果。輸出數(shù)據(jù)的格式為標(biāo)準(zhǔn)的流式數(shù)據(jù)格式FCS3.1或.BMP的圖像輸出格式,結(jié)果數(shù)據(jù)可以以散點圖及直方圖(如圖7)的形式表現(xiàn)出來,并可輕松用手觸摸。
二、 程序化實驗
MoxiFlow將較為常用的流式實驗程序化為軟件中的多個選項 (如圖8,并在不斷更新升級中),在實驗室過程中只需點擊要進行的實驗名稱即可完成相應(yīng)的數(shù)據(jù)檢測,如: 細胞活性檢測、 細胞凋亡檢、 多參數(shù)分析、細胞大小檢測、熒光珠子檢測、 細胞周期檢測(coming soon)等,使用系統(tǒng)內(nèi)相應(yīng)實驗程序即可,應(yīng)用哪里,點哪里,無需再對電壓、熒光補償?shù)葏?shù)進行調(diào)整。沒有經(jīng)過任何專業(yè)培訓(xùn)的人員也可輕松操作完成,可見MoxiFlow解放了科研人員對于流式細胞儀使用操作的繁瑣性及專業(yè)性,是對流式細胞儀的一次嶄新的**。
性 能 表 現(xiàn)
為了展現(xiàn)出MoxiFlow優(yōu)良的檢測性能,下面分別將MoxiFlow與臺式流式細胞儀、成像細胞分析系統(tǒng)、血細胞計數(shù)板進行比較。結(jié)果發(fā)現(xiàn),對于細胞的計數(shù)誤差要低于其他類型產(chǎn)品,而計數(shù)平均值的變異系數(shù)(CV)也是*小的,可見 MoxiFlow對于細胞數(shù)量的檢測表現(xiàn)優(yōu)良(圖9 左);對于細胞活性的檢測誤差值與平均活性檢測的波動性(CV)都是*小的,即在對細胞活性檢測的實驗中,MoxiFlow依然很好地保證了數(shù)據(jù)的精準(zhǔn)度和穩(wěn)定性(圖9 右)。
下圖是MoxiFlow細胞凋亡實驗的結(jié)果圖,此實驗是用喜樹堿(一種****)對Jurkat 細胞(白血病細胞)分別刺激不同時間(2h,6h,7h)所得到的散點圖??梢栽u測出該**對Jurkat細胞凋亡的作用,并給出各個群落的細胞數(shù)量、細胞濃度、占細胞總數(shù)的百分比及細胞的平均直徑等信息。
總結(jié):
外形的小巧并沒有影響MoxiFlow的應(yīng)用性能,所謂,麻雀雖小五臟俱全。它可幫你擺脫對流式細胞儀“想愛不敢愛”的矛盾心理,讓流式不再高不可攀,讓細胞在你的掌中流動。

How It Works

Moxi Z's unique, patent-protected thin-film cell count cassettes provide highly accurate, repeatable cell counts and cell size analysis in under 8 seconds (15 seconds for Type S cassette). Inspired by the gold-standard Coulter Principle used in high-end counting systems worldwide, the Moxi cassette flows thousands of cells per test through a cell sensing zone to accurately capture the impedance-based volumetric measurement of each individual cell in the sample. Impedance-based cell counting of individual cells is proven to be over 95% accurate versus a 75% accuracy with image-based cell counting methods.
No system contamination and integrated pre-filter for clogging prevention.
Moxi cassettes are designed with a unique "cell sieve" to minimize cell clumping and clogging. Add 1.5ml Accutase per 25cm2 of flask area c. incubate at RT or 37C until cells detach (typically 5-15min) d. Pipette triturate to break apart clusters 3. For suspension cell aggregates: a. Pellet cells at 300xg, 5min b. Resuspend pellet in 1-2ml Accutase c. Incubate 15min d. Pipette triturate to break apart clusters 4. Count cells on the Moxi Z, Moxi Flow, or Zepi Flow (be sure to dilute samples into standard operating range of specific cassette, if they are above the stated concentration limits)
